Functional Differences between Periportal and Perivenous Kupffer Cells Isolated by Digitonin-Collagenase Perfusion

چکیده
Introduction Liver injury commonly appears in a zonated fashion within the liver acinus. Thus many hepatotoxins primarily damage the perivenous (centrilobular, PV) region [1]. It is now well established that Kupffer cells (KCs) play a crucial role as initiators and mediators of the inflammatory process in the liver through the release of inflammatory and immunomodulatory mediators. However, the role of KCs in initiating regional damage is not established. Several studies based on immunohistochemistry have shown that the KCs in the periportal (PP) area are larger and cell size separation based on elutriation suggest that they are more active in phagocytosis [2-5]. The evidence suggesting that the smaller KCs in the centrilobular region would be more active in cytokine production is not clear-cut [3,4].

Digitonin-collagenase perfusion for efficient separation of periportal or perivenous hepatocytes.
Intact rat liver cells from the perivenous region were isolated by collagenase perfusion after first destroying the periportal region by a brief portal infusion of digitonin. Periportal cells were isolated after retrograde digitonin infusion. Significantly higher alanine aminotransferase, gamma-glutamyltransferase and lactate dehydrogenase activities and lower glutamate dehydrogenase and pyruva...
متن کاملUrea synthesis in freshly isolated and in cultured periportal and perivenous hepatocytes.
Periportal hepatocytes isolated by digitonin/collagenase perfusion produced urea faster than did similarly prepared perivenous hepatocytes, in both the presence and the absence of amino acids and various urea precursors. There was no difference between the two cell types in rates of intracellular proteolysis. The initial difference in urea synthesis persisted for 5 days during primary culture, ...

The zonal distribution of GSH metabolism was investigated by comparing hepatocytes obtained from the periportal (zone 1) or perivenous (zone 3) region by digitonin/collagenase perfusion. Freshly isolated periportal and perivenous cells had similar viability (dye exclusion, lactate dehydrogenase leakage and ATP content) and GSH content (2.4 and 2.7 mumol/g respectively). During incubation, perip...
متن کاملZonal differences in DNA synthesis and in transglutaminase activity between perivenous versus periportal regions of regenerating rat liver.
We investigated a relationship within zonal differences in DNA synthesis and in transglutaminase (TGase) activity between perivenous versus periportal regions of regenerating rat liver. Using the digitonin/collagenase perfusion technique, hepatocyte subpopulations were isolated from each region at various time points after partial hepatectomy. The amounts of DNA synthesis as well as the levels ...
متن کاملDigitonin perfusion in the study of metabolic zonation of the rat liver: potassium as an intracellular concentration reference.
The mammalian liver microcirculation seems to be uniquely organized in order to create maximum difference of the biochemical milieu to which the upstream (periportal) and downstream (perivenous) parenchymal cells are exposed. Indeed the existence of a subdivision of hepatocytes in zones of different metabolic function, long predicted by enzyme zonation studies (for a review, see Jungermann & Ka...
